Endemic spotted fever group rickettsiosis was reported in Shimane Prefecture, Japan. From an analysis of 14 clinical cases found in the endemic area, the infectious agent of spotted fever group rickettsiosis was identified as Rickettsia japonica. In this study, we also found that Rickettsia japonica was highly infected with the vector tick, Haemaphysalis longicornis, in the endemic area. These findings suggest that the high incidence of rickettsiosis in Shimane Prefecture can be explained by the high prevalence of Rickettsia japonica among Haemaphysalis longicornis ticks. 相似文献

We selectively assessed the thermal and hygrothermal treatment times of duplex heat-treated samples from the softwood hinoki cypress (Chamaecyparis obtusa) and the hardwood Japanese zelkova (Zelkova serrata) using near-infrared (NIR) spectroscopy with principal component analysis (PCA) and spectral-kinetic analysis. Wood samples from each species were thermally or hygrothermally treated at 120, 130, 150, and 180 °C, and the second-derivative spectra of these samples in the 6300–5450 cm?1 range, where moisture content has the smallest effect, were then subjected to PCA. The master curve that was calculated by kinetic analysis successfully explained changes in the first principal component (PC1) scores with thermal treatment time for all temperatures. The angles between the PC1 loadings that explained the spectral variation due to thermal and hygrothermal treatment were 79° for hinoki and 80° for zelkova. Thus, calculation of the inner product between the second-derivative spectra of duplex heat-treated wood and a loading vector that explained the spectral variation due to thermal or hygrothermal treatment allowed us to selectively assess the thermal and hygrothermal treatment times. 相似文献

Here, we evaluated the application of near-infrared (NIR) spectroscopy for estimating the degradation level of archeological wood samples from the Tohyamago area, the dendrochronological ages of which were also determined. The wood samples were radially cut from three logs obtained from the Tohyamago area. NIR reflectance spectra were measured from the tangential faces of air- and oven-dried wood samples using a Fourier transform NIR spectrophotometer. The second derivative spectra within the wavenumber range of 6400–5200 cm?1, in which the effect of moisture content in wood is suspected to be insignificant, showed a characteristic behavior with age. By comparing the second derivative spectral change in our wood samples with that in wood degraded by aging, thermal treatment, fungal attack, and lightning reported in the literature, we found that the second derivative spectra of wood samples from one log was similar to those of wood degraded by hygro-thermal treatment, whereas those of wood samples from another log was similar to those of wood degraded by brown-rot fungi. The physical and chemical properties of archeological wood were well predicted using a combination of partial least square regression analysis and NIR spectroscopy. 相似文献

Many polypeptides isolated from shallow water cnidarian species have been utilized as valuable biochemical tools in both basic and applied biological sciences. Deepwater cnidarian species might be another potential resource for novel biochemical tools. However, because of limited access to cnidarian samples from deep-sea environments, bioactive polypeptides have never before been reported from this group. In this study, we collected twelve deep-sea jellyfish species (nine hydrozoans and three scyphozoans) using a plankton net that was specially designed for collecting deep-sea organisms, and prepared water-soluble extracts, presumably containing polypeptides, of these jellyfishes. The extracts were subjected to cytotoxicity, hemolytic activity, and crustacean lethal toxicity tests. In the cytotoxicity test, six out of the nine tested hydrozoan species showed activity. In the hemolytic activity test, only three hydrozoans showed activity and none of the scyphozoan jellyfishes showed activity. In the crustacean lethality test, two hydrozoan jellyfishes and all three of the tested scyphozoan jellyfishes showed lethal activity. These results revealed a high incidence of water-soluble bioactive substances occurring in these deep-sea jellyfishes. Furthermore, all the heat-treated and the methanol-treated crude jellyfish extracts lost their bioactivities. Thus, it is likely that the bioactive compounds in the water-soluble extracts were unstable polypeptides (proteins). The disease caused by cyprinid herpesvirus-3 (CyHV-3) severely impacts the natural freshwater ecosystem and damages carp and koi farming, however, the pathway of CyHV-3 transmission remains unclear. It is possible that the virus adheres to plankton, which then facilitate viral movement and transmission, and therefore, it is hypothesised that plankton are involved in the disease dynamics. In this study, plankton were collected at eight sites in the Iba-naiko lagoon; we detected and quantified CyHV-3 DNA from plankton samples. The results of the correlation analysis showed a significant positive correlation between CyHV-3 copies and the number of Rotifera, suggesting that CyHV-3 binds to and/or is concentrated by Rotifera. Our results suggest that plankton affect viral ecology in the natural environment. 相似文献

Type Ic supernovae, the explosions after the core collapse of massive stars that have previously lost their hydrogen and helium envelopes, are particularly interesting because of their link with long-duration gamma ray bursts. Although indications exist that these explosions are aspherical, direct evidence has been missing. Late-time observations of supernova SN 2003jd, a luminous type Ic supernova, provide such evidence. Recent Subaru and Keck spectra reveal double-peaked profiles in the nebular lines of neutral oxygen and magnesium. These profiles are different from those of known type Ic supernovae, with or without a gamma ray burst, and they can be understood if SN 2003jd was an aspherical axisymmetric explosion viewed from near the equatorial plane. If SN 2003jd was associated with a gamma ray burst, we missed the burst because it was pointing away from us. 相似文献

ABSTRACT: An acidic serine carboxypeptidase (CPase Tpa) from the hepatopancreas of Japanese common squid Todarodes pacificus was purified. Purified CPase Tpa had a molecular mass of 36 kDa on sodium dodecylsulfate–polyacrylamide gel electrophoresis, and an isoelectric point of 6.0. The optimum pH of CPase Tpa was pH 4.0. In investigating the specificity of CPase Tpa for several peptide substances, it was found that peptides with hydrophobic or bulky amino acid residues at the P1 position reacted well. The enzymatic activity was almost completely inhibited by p -chloromercuribenzoic acid, monoiodoacetic acid, diisopropylfluorophosphate and HgCl2 . This is the basis for its grouping in the serine carboxypeptidase family (EC 3. 4. 16. 5). The substrate specificity of CPase Tpa can be used to eliminate the bitterness of bitter peptides. In this study, the bitterness-reductive effect using bitter peptides prepared by hydrolyzing soy protein, casein and corn gluten with pepsin or trypsin was tested. The bitterness of soy peptide digested with pepsin was completely eliminated by treatment with CPase Tpa, whereas the bitterness of casein digested with trypsin and corn peptide digested with pepsin were somewhat less efficient. On the basis of these results, it is anticipated that CPase Tpa would be effective in eliminating the bitterness of some bitter peptides. 相似文献

ABSTRACT: Two feeding experiments were conducted to elucidate growth performance of tiger puffer in a 10 m3 water volume closed system. In experiment 1, 1000 fish of 3.5 g average body weight were fed tiger puffer commercial feed twice daily to apparent satiation, 6 days a week for 224 days. Sand-filtered sea water was used and no water was exchanged during the rearing period. Immediately after cutting of lower teeth at day 112, daily feed consumption decreased greatly and 60 fish died in few days. Feeding rates recovered and then decreased gradually as nitrate levels increased from 600–1048 mg N/L. Fish grew to 343 g with 91% survival rate and 87% feed efficiency. Rearing conditions of experiment 2 were similar to experiment 1, except that culture water was exchanged to maintain the nitrate level less than 600 mg N/L during the 224-day experiment. Mortality and reduction of feed consumption occurred immediately after teeth cutting as was observed in experiment 1. Significant reduction of feed intake was not found during other rearing periods. Fish of 3 g grew to 303 g with 91% survival rate and 72% feed efficiency. 相似文献

From the two species of bivalves, Calyptogena soyoae around a cold seep and Bathymodiolus septemdierum near hydrothermal vents in the sea, sterols were isolated using high-pressure liquid chromatography analysis of the lipid
fraction guided by characteristic 1H NMR signals of the sterol skeleton. The minor sterol composition of C. soyoae included 24-methylenecycloartanal, cycloeucalenol, and obutusifoliol, which are known phytosterols. From B. Septemdierum, lathosterol and cholesterol as main sterols together with more diverse sterols were obtained. The difference between these
